选取8种玉米基因型的幼胚为外植体,以改良MS培养基为基本培养基,对基因型、幼胚大小、培养基激素配比等影响因素进行分析,研究玉米杂交种高效再生体系。结果表明:各试验材料均能诱导出一定数量的愈伤组织,但不同基因型愈伤组织的诱导率、愈伤组织的胚性诱导和保持能力存在差异,具有显著的基因型依赖性。在本试验条件下,杂交种登海11号愈伤组织的诱导率、胚性性状及其保持能力均十分优秀,是一个理想的供试材料。此外,一些重要影响因素的优化,如选用1~2 mm长的幼胚,在诱导培养基中添中2~3mg/L 2,4-D以及分化培养基中添加0.5 mg/L 6-BA,可分别显著提高登海11号愈伤组织的诱导率和分化率。
The embryos of eight maize genotypes were selected as explants. The improved MS medium was used as the basic medium to analyze the influencing factors such as the genotype, the size of immature embryos and the ratio of hormone in culture medium, and to study the efficient regeneration system of maize hybrids . The results showed that a certain amount of callus could be induced in all the tested materials. However, the callus induction rate and the embryogenic induction and maintenance ability of different genotype callus were different and had significant genotype dependence. Under the conditions of this experiment, the induction rate, embryogenic traits and their holding ability of the hybrid Denghai 11 callus are all excellent, which is an ideal material for the test. In addition, optimization of some important factors such as selection of 1-2 mm long embryos, addition of 2-3 mg / L 2,4-D in the induction medium, and addition of 0.5 mg / L 6-BA to the differentiation medium , Respectively, can significantly improve Denghai 11 callus induction rate and differentiation rate.
