拉扎碱类(lazaroids)引是脂质膜过氧化作用的抑制剂,并似乎是氧自由基的清除剂。已在数种CNS疾患中广泛研究了它的一个成员梯利拉扎(tirilazad)的可能治疗作用:对脊髓创伤有高度疗效,对实验性头部损伤模型亦显示有疗效;对蛛网膜下出血的临床研究表明有较好前景,因它能减轻血管痉挛与脑梗塞,并已在欧洲数个国家被批准用于治疗此病;对各种卒中模型亦有益。本文综述了实验与临床研究结果,并系统介绍了其药动学特性。
Lazaroids are inhibitors of lipid membrane peroxidation and appear to be scavengers of oxygen free radicals. A possible therapeutic effect of one of its members, tirilazad, has been extensively studied in several CNS disorders: highly effective on spinal cord trauma and also on curative experimental head injury models; on subarachnoid hemorrhage Clinical studies have shown promising prospects because it can reduce vasospasm and cerebral infarction and has been approved in several European countries for the treatment of the disease; it is also beneficial for a variety of stroke models. This review summarizes the experimental and clinical research results and systematically introduces its pharmacokinetic properties.
