目的:调查Y染色体上的短串联重复序列DYS552基因座在山西汉族男性人群中的基因频率分布、多态性及群体分布情况,为法医学个体识别及亲子鉴定提供计算数据,对其在法医学上的应用进行可行性探讨。方法:随机抽取102例山西汉族男性无关个体静脉血500μL,用GDB中的引物进行PCR扩增,银染显色,对其选取的等位基因测序,构建等位基因分型标准物;按照ISFG原则命名各等位基因。结果:102例山西汉族人群DYS552基因座共检出7种不同的等位基因,分别为DYS552×23(0.0194)、DYS552×24(0.1105)、DYS552×25(0.3981)、DYS552×26(0.2427)、DYS552×27(0.1262)、DYS552×28(0.0874)、DYS552×29(0.0097),基因多样性为0.7523,其个人识别能力(DP)和非父排出率(PE)均为0.7523。结论:DYS552基因座具有较高的遗传多态性,在法医学及人类遗传学方面具有较高应用价值。
OBJECTIVE: To investigate the frequency distribution, polymorphism and population distribution of DYS552 locus on Y chromosome in male Han population in Shanxi Province, and to provide computational data for forensic identification and paternity testing. In the forensic science Application feasibility study. Methods: A total of 102 venous blood samples of male Han unrelated individuals from Shanxi were randomly selected. The primers of GDB were used for PCR amplification and silver staining. The selected alleles were sequenced to construct allelic typing standards. According to ISFG Principle naming each allele. Results: There were 7 different alleles in DYS552 locus of 102 Chinese Han population, including DYS552 × 23 (0.0194), DYS552 × 24 (0.1105), DYS552 × 25 (0.3981), DYS552 × 26 (0.2427) , DYS552 × 27 (0.1262), DYS552 × 28 (0.0874) and DYS552 × 29 (0.0097), respectively. The gene diversity was 0.7523, and both the DP and the non-parent PE were 0.7523. Conclusion: The DYS552 locus has high genetic polymorphism and has high value in forensic medicine and human genetics.
