目的 :探讨重组人骨形成蛋白 2 (rhBMP 2m)对环磷酰胺 (CTX)致骨髓损伤小鼠的治疗作用。方法 :18只小鼠随机分为 3组 :即CTX注射组、BMP治疗组和PBS对照组 ,每组 6只小鼠。CTX组和BMP治疗组小鼠 ,一次性腹腔注射 2 0 0mg/kgCTX建立骨髓损伤的模型。注射CTX第 2天 ,BMP治疗组每只小鼠腹腔注射 0 .5mg的rhBMP 2m开始治疗。观察 3组小鼠外周血白细胞数的变化。分别在第 5天和第 8天 ,用流式细胞仪 (FCM )检测骨髓有核细胞中DNA的含量及细胞周期的变化 ,同时进行粒单细胞集落形成 (CFU GM)细胞培养。结果 :注射CTX后第 4天 ,BMP治疗组和CTX注射组的外周血白细胞数均降至最低点 ,然后逐渐回升 ,两组相比较无显著差异 (P >0 .0 5 )。注射CTX后第 5天 ,BMP治疗组骨髓有核细胞中 ,G0 /G1期细胞的比例较CTX组明显提高 ,细胞的坏死及凋亡率明显下降 (P <0 .0 1)。第 8天 ,BMP治疗组的骨髓有核细胞数较CTX组增加明显 (P <0 .0 1)。结论 :BMP对CTX所致小鼠骨髓的损伤具有修复治疗作用
Objective: To investigate the therapeutic effect of recombinant human bone morphogenetic protein-2 (rhBMP 2m) on bone marrow-injured mice induced by cyclophosphamide (CTX). Methods: Eighteen mice were randomly divided into three groups: CTX injection group, BMP treatment group and PBS control group, 6 mice in each group. CTX group and BMP treatment group mice, a one-time intraperitoneal injection of CTX 200mg / kg bone marrow injury model. On the second day after CTX injection, each mouse in the BMP treatment group was intraperitoneally injected with 0.5 mg of rhBMP 2m to start treatment. The changes of peripheral blood leukocytes in 3 groups were observed. On day 5 and day 8, the content of DNA and the cell cycle in bone marrow nucleated cells were detected by flow cytometry (FCM). Meanwhile, CFU GM cells were cultured. Results: On the 4th day after injection of CTX, the number of peripheral blood leukocytes in BMP-treated group and CTX-injected group all dropped to the lowest point and then gradually increased. There was no significant difference between the two groups (P> 0.05). On the fifth day after injection of CTX, the proportion of G0 / G1 phase cells in BMP-treated group was significantly higher than that in CTX group, and the necrosis and apoptosis rate of cells were significantly decreased (P <0.01). On the 8th day, the number of bone marrow nucleated cells in BMP-treated group increased significantly compared with CTX group (P <0.01). Conclusion: BMP can repair the bone marrow of mice induced by CTX
