本文应用垂直板不连续系统的聚丙烯酰胺凝胶电泳(PAGE)法,分析了5个杂交一代种(佳粉一号、佳粉二号、佳粉十号、佳粉十五号、双抗二号)发芽种苗的五种同工酶(苹果酸脱氢酶(MDH)、乙醇脱氢酶(ADH)、磷酸葡萄糖变位酶(PGM)、酯酶(EST)、异柠檬酸脱氢酶(IDH))。发现5个杂交一代种中,3个杂交一代种与其双亲的苹果酸脱氢酶(MDH)谱带有明显稳定的区别。两个杂交一代种与其双亲的酯酶(EST)谱带有区别。在此基础上,建立了用苹果酸脱氢酶电泳分析检测番茄一代杂交种纯度的技术,检测结果与田间鉴定结果基本一致。
In this paper, the polyacrylamide gel electrophoresis (PAGE) method of vertical plate discontinuous system was used to analyze five hybrid F1 hybrids (Jiafen No.1, Jiafen2, Jiafen10, Jiafen15, (MDH), alcohol dehydrogenase (ADH), phosphoglucomutase (PGM), esterase (EST), isocitrate dehydrogenation Enzyme (IDH)). Three out of five crossbreds were found to have a clearly stable difference from the malate dehydrogenase (MDH) band of their parents. The two hybrids differ from the esterase (EST) spectrum of their parents. On this basis, the technology of malate dehydrogenase electrophoresis analysis to detect the purity of tomato hybrids was established. The results of the test were basically the same as those in the field.
