用玉米粒培养基制黑木耳原种,方法简便,发菌快,能在较短时间内获得黑木耳原种,但其菌丝细弱,易老化。为克服这一不足,笔者采用木屑与玉米粒混合培养基制黑木耳原种,效果比较理想。一、材料与方法(一)供试材料菌种为丹林1号黑木耳,是本所选育。培养基配方设三个处理:A.玉米粒70%,阔叶木屑25%,麸皮4.4%,石膏0.6%;B.玉米粒99.5%,石膏0.5%;C.对照,常规木屑培养基。(二)方法步骤将籽粒饱满、无霉变的玉米粒,用40℃温水浸泡24小时再煮至种皮不破为准,捞起摊开,冷却到25℃以下时,加0.35%的石膏拌匀,同时将木屑、麦麸与剩下的0.25%石膏拌匀,加清水调含水量至60%,再将这两部分混匀后装瓶,处理B 的玉米粒处理方法同A;处理C按常规配制。三种培养基分别装于同一规格的酒瓶中,装料高15cm,于1.5kg/cm~2下灭菌1小时,接种后置26℃下培养,观察菌丝生长情况,满瓶后分别挑取长满菌丝的玉米粒接于PDA 琼脂平板中央,置26℃培养。
