目的为保证保健食品的安全性,用高效液相色谱法测定脂溶性维生素软胶囊中7种人工合成色素。方法样品经石油醚萃取、乙醇-氨水-水溶液(721)溶解超声提取后,经过WAX固相萃取柱提取净化。以甲醇和乙酸铵(0.02mol/L)为流动相,梯度洗脱法经C18色谱柱分离,保留时间和光谱图定性,外标法峰面积定量。结果该法各组分检出限为0.12~0.28mg/kg,在0.5~50.0mg/L浓度范围内呈良好的线性关系,各色素组分相关系数0.9994~0.9998。样品加标回收率为86.3%~96.8%,RSD为0.9%~4.7%。结论该方法操作简便、准确可靠,能用于保健食品脂溶性维生素软胶囊中7种合成色素的检测。
Objective To ensure the safety of health food, the seven kinds of synthetic pigments in fat - soluble vitamin soft capsules were determined by high performance liquid chromatography. Methods The samples were extracted by petroleum ether, extracted with ethanol - aqueous ammonia solution (7 2 1), extracted by WAX solid phase extraction and purified. Methanol and ammonium acetate (0.02mol / L) as the mobile phase, gradient elution C18 column separation, retention time and spectroscopy qualitative, external standard peak area quantitative. Results The limit of detection of each component was 0.12 ~ 0.28mg / kg. The linear range was 0.5 ~ 50.0mg / L. The correlation coefficient of each pigment component was 0.9994 ~ 0.9998. The spiked recoveries of samples ranged from 86.3% to 96.8% with RSD of 0.9% -4.7%. Conclusion The method is simple, accurate and reliable and can be used for the determination of seven synthetic pigments in the fat-soluble vitamin soft capsule of health food.
