目的研究胃癌及增生性息肉组织中核因子κB(NF-κB)的DNA结合活性,NF-κBp65及环氧合酶2(COX-2)的表达,通过比较两者表达的差异,从而探讨NF-κB及COX-2在胃癌病程中的意义。方法收集65例外科手术胃癌及癌旁组织标本,48例消化内镜下增生性息肉组织标本。采用凝胶迁移滞留试验EMSA检测其NF-κBDNA结合活性,免疫组化SABC法测定其NF-κBp65及COX-2蛋白表达水平。结果胃癌标本NF-κBDNA结合活性更高,可见明显核移位现象,与癌旁组织及增生性息肉组织比较具有统计学差异(P<0.01),NF-κBp65在胃癌组织,癌旁组织及增生性息肉中表达阳性率分别为87.7%、10.8%和6.3%,COX-2在胃癌组织,癌旁组织及胃炎中表达阳性率分别为100%,12.3%和10.4%。相关性分析显示胃癌组织中NF-κBp65和COX-2表达呈明显正相关(r=0.6,P<0.01)。结论NF-κBDNA结合活性及NF-κBp65和COX-2在胃癌组织中阳性表达及DNA显著高于癌旁组织和增生性息肉组织,两者存在明显正相关,推测可能通过干预其活性和表达对胃癌的发生发展过程研究有一定帮助。
Objective To investigate the DNA-binding activity of NF-κB and the expressions of NF-κBp65 and COX-2 in gastric cancer and hyperplastic polyps, and to explore the difference between the expression of NF- The significance of κB and COX-2 in the course of gastric cancer. Methods Totally 65 cases of surgical specimens of gastric cancer and paracancerous tissues and 48 cases of proliferative polyp tissues were collected. EMSA was used to detect the NF-κB DNA binding activity. The expression of NF-κBp65 and COX-2 protein was detected by immunohistochemical SABC method. Results The NF-κB DNA binding activity of gastric cancer specimens was higher, showing obvious nuclear translocation. Compared with the adjacent tissues and proliferative polyps, there was a significant difference (P <0.01). NF-κBp65 expression in gastric cancer tissues, paracancerous tissues and hyperplasia The positive expression rates of COX-2 in gastric cancer tissue, pericarcinoma tissue and gastritis were 87.7%, 10.8% and 6.3% respectively, and the positive rates were 100%, 12.3% and 10.4% respectively. Correlation analysis showed that the expressions of NF-κBp65 and COX-2 in gastric cancer tissues were positively correlated (r = 0.6, P <0.01).
