目的:比较以聚鸟氨酸、聚赖氨酸为囊材制备的新生猪胰岛细胞微囊的体内、体外分泌功能差异。方法:于高压静电场中用聚电解质络合法分别制备包裹新生猪胰岛细胞的海藻酸钠-聚赖氨酸-海藻酸钠(APA)与海藻酸钠-聚鸟氨酸-海藻酸钠微囊(ALA),行体外培养及糖尿病大鼠腹腔内移植。结果:2组新生猪胰岛细胞微囊的培养液中胰岛素水平均以第3天为最高,组间差异无显著性;胰岛素刺激释放试验组间差异无显著性;体内移植后,ALA组较APA组降血糖作用稍强。结论:APA组、ALA组体外分泌功能相似,体内移植后ALA组降糖作用略强。
OBJECTIVE: To compare the in vitro and in vivo secretory functions of neonatal porcine islet microcapsules prepared from polyornithine and polylysine. Methods: Sodium alginate-polylysine-sodium alginate (APA) and sodium alginate-polyornithine-sodium alginate microbe encapsulating neonatal porcine islet cells were prepared by polyelectrolyte complexation in high voltage electrostatic field. Bladder (ALA), in vitro culture and diabetic rat intraperitoneal transplantation. Results: Insulin levels in culture medium of 2 groups of newborn porcine pancreatic islet cells were the highest on day 3 with no significant difference between groups. There was no significant difference between groups in insulin stimulation and release test after transplantation. Compared with APA Group hypoglycemic effect slightly stronger. Conclusion: The secretory function of APA group and ALA group is similar in vitro. The hypoglycemic effect of ALA group is slightly stronger after transplantation.
