胃癌组织KLF8高表达及其临床意义

目的探讨转录因子KLF8在胃癌组织中的表达及其临床意义。方法应用免疫组织化学SP法和RT-PCR法检测胃癌组织及正常胃壁组织中KLF8的表达。结果 RT-PCR法显示胃癌组织与正常胃组织中均有KLF8基因表达,正常胃组织mRNA相对表达量为0.23±0.055,胃癌组织为0.57±0.041,差异具有统计学意义(P<0.001)。免疫组织化学显示30例正常胃组织中KLF8阳性表达率为26.67%(8/30),50例胃癌组织中阳性率为49.12%(28/57),且表达与肿瘤浸润深度和有无淋巴结转移有关,与患者性别、年龄、肿瘤分化程度及TNM分期无关,并随着浸润深度的增加和淋巴结转移,KLF8表达呈上升趋势。结论 KLF8高表达与胃癌发生发展及局部侵袭转移有关,可作为胃癌诊断及预后评估的客观指标。 Objective To investigate the expression of KLF8 in gastric cancer and its clinical significance. Methods Immunohistochemical SP method and RT-PCR method were used to detect the expression of KLF8 in gastric cancer tissue and normal gastric tissue. Results The expression of KLF8 gene in gastric cancer tissue and normal gastric tissue was detected by RT-PCR. The relative expression of normal gastric tissue mRNA was 0.23 ± 0.055 and that of gastric cancer tissue was 0.57 ± 0.041, with statistical significance (P <0.001). Immunohistochemistry showed that the positive expression rate of KLF8 in 30 normal gastric tissues was 26.67% (8/30), the positive rate was 49.12% (28/57) in 50 gastric carcinoma tissues, and the positive rate of KLF8 expression was related to the depth of tumor invasion and lymph node metastasis Which was not related to patient’s gender, age, tumor differentiation and TNM stage. With the increase of invasion depth and lymph node metastasis, the expression of KLF8 increased. Conclusion The high expression of KLF8 is associated with the development of gastric carcinoma and local invasion and metastasis, which can be used as an objective indicator for the diagnosis and prognosis of gastric cancer.
