目的:了解2008年-2009年东莞市H1N1流感病毒血凝素HA1基因的特征及变化。方法:采用RT-PCR、基因测序等技术得到标本HA1基因的核酸序列,利用生物信息软件对核酸及氨基酸的特征及变化情况进行分析。结果:两年间有16个氨基酸位点发生变异,6个发生在抗原决定簇,Ca区2个,Sb区4个。受体结合位点有4个变异,130环1个,190螺旋3个。潜在糖基化位点及二硫键比较稳定,2年间没有发生变化。结论:2008年-2009年两年间H1N1流感病毒血凝素HA1基因氨基酸替换速率较高。
Objective: To understand the characteristics and changes of HA1 gene of H1N1 influenza virus in Dongguan from 2008 to 2009. Methods: The nucleic acid sequence of HA1 gene was obtained by RT-PCR and gene sequencing. The characteristics and changes of nucleic acids and amino acids were analyzed by bioinformatics software. RESULTS: Sixteen amino acid sites were mutated in two years, with six in the antigenic determinant, two in the Ca region and four in the Sb region. Receptor binding sites have 4 mutations, 130 ring 1, 190 3 spiral. Potential glycosylation sites and disulfide bonds were relatively stable, unchanged for 2 years. Conclusion: The amino acid substitution rate of HA1 gene of H1N1 influenza virus was high between 2008 and 2009.
