目的明确乙型肝炎病毒(HBV)前C第17位和29位密码子突变体对e抗原合成和分泌的影响。方法用PCR 方法扩增HBV前C/C基因区,克隆后测序及进行序列分析。将HBV前C区T1862/A1899位点突变体亚克隆人表达 质粒 pGEMT中,经体外翻译后,比较野毒株和变异株的表达产物。结果该前 C区突变株在体外仍能合成 HBV e前体 蛋白。结论前 C区 T1862/A1899突变并不能阻断 HBV e抗原前体蛋白合成,e抗原前体可能在分泌过程中受阻。
Objective To clarify the effect of pre-C codon 17 and codon 29 mutations on the synthesis and secretion of e antigen in Hepatitis B virus (HBV). Methods The pre-HBV C / C gene region was amplified by PCR, cloned, sequenced and sequenced. The T1862 / A1899 site mutants in the pre-HBV C region were subcloned into the expression vector pGEMT. After translation in vitro, the expression products of wild-type and mutant strains were compared. Results The pre-C mutant still could synthesize HBV e precursor protein in vitro. Conclusion The mutation of T1862 / A1899 in pre-C region can not block the synthesis of HBV e antigen precursor protein. The precursor of e antigen may be blocked in the process of secretion.
