改进式海登氏苏木精法,是李振岐等(1985)通过对原海登汉氏苏木精法加以改进而探索出的一种真菌细胞核染色方法.实验证明,该方法对许多植物病原真菌的细胞核具有良好的染色效果,但关于该方法对食用真菌担孢子的核染色效果,国内外尚未见报道.我们于1989年1~12月结合食用真菌细胞核相研究的需要,进行了这方面的试验.结果表明,如果将该方法直接用于食用菌担孢子细胞核染色,效果不好,若作些改进则效果大为改观.现将改进染色的技术简报如下.
