目的:建立高效液相色谱法同时测定妇炎丸中绿原酸、栀子苷、芍药苷、橙皮苷和连翘苷5种活性成分含量的方法。方法:采用CNW Athena C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈(A)-0.2%磷酸水溶液(B)为流动相,梯度洗脱,流速1.0 m L·min-1,柱温30℃,检测波长:217 nm(绿原酸、橙皮苷和连翘苷)、230 nm(栀子苷和芍药苷)。结果:绿原酸、栀子苷、芍药苷、橙皮苷和连翘苷的线性范围分别为8.80~220μg·m L-1(r=0.999 8)、4.28~107μg·m L-1(r=0.999 9)、4.16~104μg·m L-1(r=0.999 9)、8.08~202μg·m L-1(r=0.999 7)和4.52~113μg·m L-1(r=0.999 8);平均加样回收率(n=6)分别为98.9%、99.1%、98.9%、99.3%和99.2%,RSD分别为1.0%、0.9%、1.1%、1.0%和0.9%;含量分别为0.88、0.41、0.35、0.98和0.38 mg·g-1。结论:本法专属性强,结果准确,重复性好,可用于妇炎丸的质量控制。
Objective: To establish a method for the simultaneous determination of five active ingredients of chlorogenic acid, geniposide, paeoniflorin, hesperidin and forsythin in Fuyan Pill by high performance liquid chromatography. Methods: A CNW Athena C18 column (4.6 mm × 250 mm, 5 μm) was used as the mobile phase. The mobile phase was acetonitrile (A) -0.2% phosphoric acid solution (B) 30 ℃, detection wavelength: 217 nm (chlorogenic acid, hesperidin and forsythin), 230 nm (geniposide and paeoniflorin). Results: The linear ranges of chlorogenic acid, geniposide, paeoniflorin, hesperidin and forsythin were 8.80 ~ 220μg · m L -1 (r = 0.999 8), 4.28 ~ 107μg · m L -1 (r = 0.999 9), 4.16-104μg · m L -1 (r = 0.999 9), 8.08-220μg · m L -1 (r = 0.999 7) and 4.52-113μg · m L -1 (r = 0.999 8) The average recoveries (n = 6) were 98.9%, 99.1%, 98.9%, 99.3% and 99.2%, respectively, with RSDs of 1.0%, 0.9%, 1.1%, 1.0% and 0.9% 0.41, 0.35, 0.98 and 0.38 mg · g -1. Conclusion: This method is specific, accurate, reproducible and can be used for the quality control of Fuyan Pills.
