目的从侧群(side population,SP)表型细胞具有干细胞特性的角度出发,对肝癌细胞系中的SP细胞进行检测、分离和评价。方法利用SP细胞能将荧光染料Hoechst33342泵到细胞外的特性,结合流式细胞技术进行分离纯化。结果在肝癌细胞系PLC、Huh7、Hep3B、SMMC7721及1例临床肝癌组织中检测到SP细胞的比例依次为0.8%、0.3%、0.6%、0.3%、0.4%;未在HepG2中检测到SP细胞。NOD-SCID荷瘤实验发现SP细胞较非SP细胞具有更高的成瘤性。结论肝癌细胞系及标本组织含有少量的SP细胞,这些SP细胞比非SP细胞具有更强的成瘤性,为进一步分析SP细胞的肿瘤干细胞特性奠定了基础。
OBJECTIVE To detect, isolate and evaluate SP cells in hepatoma cell lines from the perspective of stem cell characteristics of side population (SP) phenotype cells. Methods SP cells were used to pump the fluorescent dye Hoechst 33342 into the extracellular space and purified by flow cytometry. Results The percentage of SP cells detected in hepatocellular carcinoma cell lines PLC, Huh7, Hep3B, SMMC7721 and one case of clinical liver cancer were 0.8%, 0.3%, 0.6%, 0.3% and 0.4%, respectively. SP cells were not detected in HepG2 . NOD-SCID tumor-bearing experiments found that SP cells have higher tumorigenicity than non-SP cells. Conclusion The hepatocellular carcinoma cell lines and tissues contain a small amount of SP cells. These SP cells have stronger tumorigenicity than non-SP cells, which lays the foundation for further analysis of tumor stem cell characteristics of SP cells.
