目的 探讨碘缺乏 (低碘 )、甲状腺功能减退 (甲减 )大鼠仔鼠海马组织c -fos、c -jun的表达。方法 分别选用低碘饲料及他巴唑诱导建立低碘及甲减大鼠仔鼠动物模型 ,取生后 2 0、3 0d仔鼠海马免疫组织化学S -P法染色 ,对c -fos、c -jun表达产物进行图像分析。结果 生后 2 0d低碘、甲减组仔鼠海马组织c -fos灰度值分别为 162 .76± 1.0 8、15 9.80± 3 .85 ,显著高于对照组 15 0 .91± 4.95 (P <0 .0 1) ;c -jun灰度值分别为 14 6.87± 1.97、14 0 .2 7± 12 .11,也显著高于对照组 13 0 .71± 8.73 (P <0 .0 1)。生后 3 0d低碘、甲减组仔鼠海马组织c -fos灰度值分别为 162 .98± 0 .83、163 .2 9±1.5 2 ,显著高于对照组 15 3 .3 2± 4.70 (P <0 .0 1) ;c -jun灰度值分别为 14 5 .80± 1.3 9、14 6.5 7± 1.5 1,明显高于对照组12 5 .2 9± 3 .5 1(P <0 .0 1)。结论 碘缺乏、甲状腺功能减退可降低仔鼠海马组织原癌基因c -fos、c -jun表达 ,进而影响神经系统及智力发育。
Objective To investigate the expression of c-fos and c-jun in hippocampus of neonatal rats with iodine deficiency (hypothyroidism) and hypothyroidism (hypothyroidism). Methods Low iodine diet and methimazole were used to establish animal model of hypodermic hypothyroidism and hypothyroidism, respectively. Immunohistochemical S-P staining was used to detect the expression of c-fos, c -jun expression product for image analysis. Results The gray value of c-fos hippocampus of pups in hypodermic and hypothyroidism groups at 20 days after birth was 162.76 ± 1.08,15.980 ± 3.85, which was significantly higher than that of control group <0.01). The gray values of c-jun were 14 6.87 ± 1.97 and 14 0 2 7 ± 12.11, respectively, which were also significantly higher than that of the control group 13 0 .71 ± 8.73 (P <0 01) . The gray values of c-fos in hippocampus of pups with hypocholesterolemia and hypothyroidism on the 30th day after birth were 162.98 ± 0.83,163.29 ± 1.52, which were significantly higher than those of the control group 15.3.3 ± 4.70 (P <0.01). The gray value of c-jun was 14.580 ± 1.3 9,146.57 ± 1.51 respectively, which was significantly higher than that of the control group 12.5.29 ± 3.51 (P < 0 .0 1). Conclusions Iodine deficiency and hypothyroidism can reduce the expression of c-fos and c-jun oncogene in the hippocampus of the offspring, which may affect the nervous system and mental development.
