目的:研究人急性淋巴细胞白血病细胞系(Molt4细胞)容积激活性氯电流。方法:采用膜片钳全细胞方式记录细胞外低渗液(160mOsmol/L)激活的Molt4细胞容积激活性氯电流,分析该电流的特性。结果:Molt4细胞在等渗溶液中背景电流较小且稳定,低渗溶液诱导激活容积激活性氯电流,该电流呈现较明显的外向优势,在钳制电压0mV~±120mV及脉冲波宽200ms条件下,没有观察到明显的电压依赖性失活和时间依赖性失活。该电流具有容积敏感性,可被细胞外高渗刺激所抑制。氯通道阻断剂他莫昔芬显著抑制容积激活性氯电流,对内向电流和外向电流的抑制作用无显著差异。结论:Molt4细胞膜存在容积敏感性氯通道;低渗刺激可激活该通道产生容积激活性氯电流;该通道对氯通道阻断剂他莫昔芬敏感。
Objective: To study the volume-activated chloride current of human acute lymphoblastic leukemia cell line (Molt4). Methods: The volume-activated chloride current of Molt4 cells activated by extracellular hypotonic solution (160mOsmol / L) was recorded by patch-clamp whole cell method and the characteristics of the current were analyzed. Results: The background current of Molt4 cells in isotonic solution was small and stable, and the hypotonic solution induced the activation of volume-activated chloride current. The current showed a significant outward-oriented advantage. Under the conditions of clamping voltage of 0mV ~ ± 120mV and pulse wave width of 200ms , No significant voltage-dependent inactivation and time-dependent inactivation were observed. This current is volume sensitive and can be inhibited by extracellular hypertonic stimuli. Chloride channel blocker tamoxifen significantly inhibited the volume-activated chloride current, the inward current and outward current inhibition was no significant difference. CONCLUSION: There is volume-sensitive chloride channel in Molt4 cell membrane. Hypotonic stimulation activates this channel to generate volume-activated chloride current. This channel is sensitive to chloride channel blocker tamoxifen.
