目的 :探讨超低浓度的丝裂霉素在大肠癌细胞光动力学疗法增效作用中对癌细胞杀伤作用规律。方法 :在癌细胞培养的基础上 ,用超低浓度的丝裂霉素增效 ;光动力学疗法后采用荧光光度法和MTT法对癌细胞内光敏剂和细胞存活率进行检测 ,数据经SPLM软件包处理。结果 :超低浓度的丝裂霉素可使癌细胞内光敏剂显著增加 ;细胞内光敏剂含量与激光照射后细胞存活率呈显著负相关 ;光动力学疗法后癌细胞存活率曲线明显左移 ,在 8、16小时细胞存活率显著降低 ;癌细胞与光敏剂作用 8~ 16小时激光照射可达到最佳杀伤效果。结论 :单独超低浓度的丝裂霉素对癌细胞无杀伤作用 ,可通过增加癌细胞内光敏剂含量来达到增效作用 ,给药时间和杀伤作用有明显的规律性。
Objective : To investigate the ultra-low concentration of mitomycin in the cytotoxicity of colorectal cancer cells in photodynamic therapy. Methods: On the basis of cancer cell culture, ultra-low concentration of mitomycin was used to synergize; after photodynamic therapy, photosensitizer and cell survival rate in cancer cells were detected by fluorescence spectrometry and MTT assay. The data was analyzed by SPLM. Package processing. RESULTS: Ultra-low concentration of mitomycin could significantly increase the photosensitizer in cancer cells; the content of photosensitizer in cells was significantly negatively correlated with the survival rate of cells after laser irradiation; the survival rate curve of cancer cells significantly shifted left after photodynamic therapy. At 8 and 16 hours, the cell viability was significantly reduced; the effect of cancer cells and the photosensitizer on the laser irradiation for 8 to 16 hours can achieve the best killing effect. Conclusion: Ultra-low concentration of mitomycin alone has no killing effect on cancer cells. It can increase synergistic effect by increasing the content of photosensitizer in cancer cells. The administration time and killing effect have obvious regularity.
