目的 探讨树突状细胞(DC)在肾切除大鼠肾小管间质中的分布与作用及缬沙坦对DC作用的影响。方法 建立慢性肾衰5/6肾切除大鼠模型,随机分为正常组(n=18)、假手术组(n=18)、模型组(n=18)和缬沙坦治疗组(n=18)。分别于建模1、4、12周,采用免疫双染和荧光图像分析法观察CD1a+CD80+DC在各组大鼠肾组织中的分布变化;免疫组化法观察P-选择素和各纤维化相关指标在肾组织中的表达;Masson染色评定各组肾小管间质纤维化(TIF)程度。结果 模型组CD1a+CD80+DC主要分布于肾小管、肾间质和肾血管,以肾间质最为明显;其分布数量于12周较1周、4周明显增多,且与慢性肾功能减退呈正相关;12周时,P-选择素和各纤维化相关指标以肾小管间质为主表达和TIF程度明显增加,并与CD1a+CD80+DC分布数量呈正相关;经缬沙坦治疗后,CD1a+CD80+DC分布减少,P-选择素和各纤维化相关指标表达下降,TIF程度减轻及肾功能改善。结论 DC可能启动或参与了肾小管间质纤维化形成,并与肾功能损害程度密切相关,缬沙坦对此具有明显的抑制和肾脏保护作用。
Objective To investigate the distribution and function of dendritic cells (DCs) in the tubulointerstitium of nephrectomized rats and the effect of valsartan on DCs. Methods A rat model of chronic renal failure 5/6 nephrectomy was established and randomly divided into normal group (n = 18), sham operation group (n = 18), model group (n = 18) and valsartan treatment group (n = 18). The changes of distribution of CD1a + CD80 + DC in renal tissues of rats in each group were observed by immunofluorescence staining and fluorescence image analysis respectively at 1, 4, 12 weeks after modeling. The expressions of P-selectin and various fibers The expression of related indicators in renal tissue; Masson staining to assess the degree of tubulointerstitial fibrosis (TIF). Results The distribution of CD1a + CD80 + DC in the model group was mainly in the renal tubules, renal interstitium and renal vessels with the most obvious in the renal interstitium. The distribution of CD1a + CD80 + DC in the model group was significantly higher than that in the 12th week, the 4th week, and positively correlated with the chronic renal failure ; At 12 weeks, the expression of P-selectin and fibrosis correlated mainly with tubulointerstitial and TIF, and positively correlated with the distribution of CD1a + CD80 + DC. After valsartan treatment, CD1a + CD80 + DC distribution decreased, P-selectin and various indicators of fibrosis decreased expression, reduce the degree of TIF and renal function improved. Conclusion DC may initiate or participate in the formation of tubulointerstitial fibrosis and is closely related to the degree of renal dysfunction. Valsartan has obvious inhibitory effect and renal protection.
