用蒽酮-硫酸法和3,5-二硝基水杨酸(DNS)法测定多糖含量。以多糖得率为指标,采用正交实验法分别对水提和醇沉过程的条件进行优选,最佳水提工艺为:提取温度100℃,料液比1:10,提取2次,每次4h;最佳醇沉工艺为:浓缩液加入3倍体积100%乙醇,沉淀4h,此工艺条件下多糖得率为1.58%。该工艺简单,适于杜仲皮多糖提取。
Determination of polysaccharide content by anthrone - sulfuric acid method and 3,5 - dinitrosalicylic acid (DNS) method. Using polysaccharide yield as an index, orthogonal test was used to optimize the conditions of water extraction and alcohol precipitation respectively. The optimum extraction conditions were: extraction temperature 100 ℃, solid-liquid ratio 1:10, extraction 2 times, 4h; the best alcohol precipitation process: concentrated solution was added 3 times the volume of 100% ethanol, precipitation 4h, polysaccharide yield of 1.58% under this process conditions. The process is simple, suitable for Eucommia bark polysaccharide extraction.
