提取高质量线粒体DNA(mtDNA)是研究大白菜细胞质雄性不育系不育分子特性的前提。以大白菜绿色叶片为试验材料,应用一种省时、简便、操作步骤少的方法,快速获得3组11份大白菜的mtDNA(包括保持系、Ogu CMS、Pol CMS和CMS96细胞质雄性不育系)。试验中不需要加入DNase I酶消化核DNA,提取的mtDNA质量较好;同时,采用RAPD技术,对11份大白菜mtDNA进行多态性分析,获得了与大白菜Ogu CMS和CMS96不育系相关的差异片段10条,与大白菜Ogu CMS不育系相关的差异片段3条。初步证实:大白菜CMS96和大白菜Ogu CMS不育系既有相似性又存在差异性。
Extracting high-quality mitochondrial DNA (mtDNA) is a prerequisite to study the molecular characteristics of cytoplasmic male sterile lines in Chinese cabbage. Using green leaves of Chinese cabbage as test material, mtDNA (including maintainer line, Ogu CMS, Pol CMS and CMS96 cytoplasmic male sterility lines) of 11 Chinese cabbages were quickly obtained by using a method that is time-saving, simple and with few operation steps. ). In the experiment, it was not necessary to add DNase I enzyme to digest nuclear DNA, and the extracted mtDNA was of good quality. Meanwhile, the polymorphism analysis of 11 Chinese cabbage mtDNAs was carried out by using RAPD technique, which was related to Ogu CMS and CMS96 CMS of Chinese cabbage 10 differential fragments and 3 differential fragments related to Ogu CMS in Chinese cabbage. Preliminary confirmed: Chinese cabbage CMS96 and Chinese cabbage Ogu CMS both similarities and differences.
