目的探讨姜黄素对人胃低分化腺癌MGC-803细胞株增殖及细胞中核因子κB(NF-κB)蛋白表达的影响。方法将培养的MGC-803细胞分为对照组(A组)、5μmol/L姜黄素作用组(B组)、10μmol/L姜黄素作用组(C组)和20μmol/L姜黄素作用组(D组),分别干预24、48、72h,用cellcounting Kit-8(CCK-8)法检测细胞增殖抑制情况,用免疫蛋白印迹(Western blot)法检测各组作用24h后细胞中NF-κB表达情况。结果作用24、48、72h,不同浓度的姜黄素对胃癌细胞MGC-803的增殖有抑制作用,且随姜黄素浓度的增加和作用时间的延长,增殖抑制率逐渐增加。Western blot法检测显示,作用24h后A、B、C、D组MGC-803中NF-κB蛋白表达量分别为(0.889±0.019)、(0.640±0.019)、(0.503±0.014)和(0.349±0.022),随着作用浓度的增加,NF-κB表达量逐渐降低,组间差异均有统计学意义(P<0.01)。结论姜黄素能够抑制胃癌细胞MGC-803增殖,且随着姜黄素浓度的增加和作用时间的延长,细胞增殖抑制率逐渐增加;胃癌细胞MGC-803存在NF-κB的表达;姜黄素通过下调NF-κB的表达从而对胃癌细胞MGC-803的增殖起到抑制作用。
Objective To investigate the effect of curcumin on the proliferation of human gastric poorly differentiated adenocarcinoma MGC-803 cells and the expression of nuclear factor κB (NF-κB) protein. Methods MGC-803 cells were divided into control group (A group), curcumin group (B group) (5μmol / L), curcumin group (C group) Group). The cell proliferation was detected by cell counting Kit-8 (CCK-8) method. The expression of NF-κB in each group was detected by Western blotting 24 h, 48 h and 72 h after transfection. . Results The effect of different concentrations of curcumin on the proliferation of gastric cancer cell line MGC-803 was inhibited at 24, 48, and 72 hours. With the increase of curcumin concentration and the prolongation of action time, the proliferation inhibition rate gradually increased. The results of Western blot showed that the expression of NF-κB in MGC-803 cells in groups A, B, C and D after 24h was (0.889 ± 0.019), (0.640 ± 0.019), (0.503 ± 0.014) and (0.349 ± 0.022). With the increase of concentration, the expression of NF-κB gradually decreased, with significant difference between the two groups (P <0.01). Conclusion Curcumin can inhibit the proliferation of gastric cancer cell line MGC-803. With the increase of curcumin concentration and prolongation of action time, curcumin inhibits the proliferation of MGC-803 cells. The expression of NF-κB in gastric cancer cell line MGC- -κB expression of gastric cancer cell MGC-803 inhibited the proliferation.
