制备了抗尿激酶血清,并由此抗血清提取抗尿激酶IgG.将此IgG偶联到CNBr活化的Sepharose4B上,对尿激酶(UK)粗品进行纯化.经SDS-聚丙烯酰胺凝胶电泳分析,纯化的UK样品显示3条蛋白带,高分子量尿激酶(H-UK),低分子量尿激酶(LUK)及分子量为1.8×10~4道尔顿的UK肽链.纯化UK样品的比活为79143~101710 IU/mg蛋白,从层析柱上的回收率为51~82%.未纯化UK粗品于SDS-聚丙烯酰胺凝胶电泳中显示9~10条带.
Anti-urokinase serum was prepared, and anti-urokinase IgG was extracted from the antisera.This IgG was coupled to CNBr-activated Sepharose 4B to purify the urokinase (UK) crude.After SDS-polyacrylamide gel electrophoresis , Purified UK samples showed 3 bands with high molecular weight urokinase (H-UK), low molecular weight urokinase (LUK), and UK peptide chains with a molecular weight of 1.8 x 10 ~ 4 Daltons. Was 79143-101710 IU / mg protein and the recovery from the column was 51-82%. Crude unpurified UK showed 9-10 bands on SDS-polyacrylamide gel electrophoresis.
