目的评价多次5-氨基乙酰丙酸(5-aminolevulinic acid,5-ALA)介导光动力下(5-ALA-PDTs)存活鼻咽癌(NPC)细胞亚株CNE2和5-8F的生物学行为。方法采用半数致死剂量(LD50)对细胞亚株CNE2和5-8F间断进行3次PDT处理,收集存活细胞传代扩增,然后分别评价细胞增殖、黏附、迁移能力和血管生成素1、2(Ang-1,Ang-2)表达的变化。结果多次5-ALA-PDTs后存活的CNE2和5-8F在增殖、粘附、迁移能力上均有下降,Ang-1和Ang-2表达下调(P<0.05)。结论多次5-ALA-PDTs可使存活NPC细胞的恶性生物学行为稳定下降,具有减少淋巴转移的潜在价值。
Objective To evaluate the biology of 5-aminolevulinic acid (5-ALA) -mediated survival of nasopharyngeal carcinoma (NPC) cell subline CNE2 and 5-8F under 5-ALA-PDTs behavior. Methods Cell subtypes CNE2 and 5-8F were treated with PDT at a dose of half the lethal dose (LD50) for 3 times. The surviving cells were collected and passaged. The cell proliferation, adhesion, migration and angiopoietin 1,2 (Ang) -1, Ang-2) expression changes. Results CNE2 and 5-8F survived after repeated 5-ALA-PDTs decreased in proliferation, adhesion and migration, and decreased Ang-1 and Ang-2 expression (P <0.05). Conclusions Multiple 5-ALA-PDTs can stably decrease the malignant biological behavior of surviving NPC cells and have the potential value of reducing lymphatic metastasis.
