目的:观察体外TNFα联用IL-2培养LAK细胞的抗胃癌作用及TNFα预处理后胃癌细胞对LAK细胞敏感性的变化。方法:以不同浓度TNFα与IL-2配伍培养LAK细胞,并做TNFα的单抗阻断试验,用LDH释放法检测LAK细胞抗癌活性;以TNFα预处理胃癌细胞,检测LAK细胞抗瘤活性。结果:TNFα在低浓度IL-2下增加LAK活性,使IL-2诱导同等LAK活性的浓度下降约10倍;TNFα预处理使LAK对其抗瘤活性提高约9%。结论:TNFα可增加IL-2诱导的LAK抗胃癌活性;TNFα预处理可使胃癌细胞对LAK的敏感性增加
OBJECTIVE: To observe the anti-cancer effect of LAK cells cultured with TNFα in combination with IL-2 in vitro and the changes of sensitivity of LAK cells to gastric cancer cells treated with TNFα. METHODS: LAK cells were cultured with different concentrations of TNFα and IL-2. LAK monoclonal antibody blocking assay was performed. The anticancer activity of LAK cells was detected by LDH release method. Gastric cancer cells were pretreated with TNFα and the antitumor activity of LAK cells was detected. RESULTS: TNFα increased LAK activity at low concentrations of IL-2, resulting in a 10-fold decrease in the concentration of IL-2 induced equivalent LAK activity; pretreatment with TNFα increased the anti-tumor activity of LAK by approximately 9%. Conclusion: TNFα can increase IL-2-induced anti-gastric activity of LAK; TNFα pretreatment can increase the sensitivity of gastric cancer cells to LAK
