Klotho基因是Kroo-o等~([1])在研究自发性高血压时发现的与人类衰老密切相关的基因。该基因与人类心血管疾病、肾脏疾病及骨质疏松等衰老性疾病密切相关。近年来的研究认为Klotho基因的异常表达可能参与到肿瘤的发生、发展,作为一个抑癌基因参与调控多种肿瘤的生物学过程~([2])。本文现就Klotho基因及其与肿瘤的关系进行综述。1 Klotho基因及蛋白的生物学特性1.1 Klotho基因结构人类的Klotho基因定位于染色体13q12,全长约50kb,包括5个外显子和4个内含子。在Klotho基因的结构中,有一个潜在的可变剪切位点存在于其mRNA上,定位于外显子3中,如果此位点接受了一个50bp片段的插入,该序列末断的两个核苷酸
