目的探讨三磷酸腺苷(ATP)在大鼠胰岛分离过程中抑制胰酶对胰岛细胞的消化作用。方法将20只SD大鼠平均分为两组,处理组在胰岛分离全过程中加用ATP进行干预,对照组则不加ATP处理,采用Ficoll非连续密度离心纯化后对两组胰岛细胞的凋亡、胰岛产量及活力方面进行比较性研究。结果加用ATP的处理组的胰岛细胞凋亡率显著低于对照组(P<0.01),处理组胰岛产量和胰岛素释放水平均显著高于对照组(P<0.01)。结论胰岛分离过程中加用一定浓度ATP对胰岛具有抑制凋亡和保护胰岛活力的作用。
Objective To investigate the inhibitory effect of adenosine triphosphate (ATP) on pancreatic islet cells digestion during islet isolation in rats. Methods Twenty SD rats were equally divided into two groups. The treatment group was treated with ATP during the whole process of islet isolation, while the control group without ATP treatment. The Ficoll non-continuous density centrifugation was used to purify the islet cells of both groups Death, islet production and vitality comparative study. Results Compared with the control group, the apoptosis rate of islet cells in the treatment group treated with ATP was significantly lower than that in the control group (P <0.01). The islet production and the release of insulin in the treatment group were significantly higher than those in the control group (P <0.01). Conclusion Addition of certain concentration of ATP during islet isolation can inhibit the islets and protect the islet activity.
