目的探讨胎盘组织中血清与糖皮质激素诱导蛋白激酶-1(SGK1)和转化生长因子β1(TGF-β1)在子痫前期发生与发展中的作用。方法选取2014年1月-2015年1月该院收治的子痫前期孕产妇79例作为研究组,其中重度子痫前期39例作为A组,轻度子痫前期40例作为B组,同期选取正常孕产妇40例作为对照组,采用免疫组化SP法检测胎盘组织SGK1、TGF-β1表达,Masson染色检测法检测胎盘组织胶原纤维沉积。比较组间胎盘组织SGK1、TGF-β1表达的差异,采用Pearson相关性分析胎盘组织SGK1、TGF-β1表达与胶原纤维沉积的关系。结果 A、B两组胎盘组织SGK1、TGF-β1阳性表达率与阳性表达量明显高于对照组,其中A组盘组织SGK1、TGF-β1上述指标明显高于B组,比较差异有统计学意义(P<0.05)。A、B两组胎盘组织胶原纤维沉积率明显高于对照组,A组上述指标明显高于B组,差异有统计学意义(P<0.05)。Pearson相关性分析胎盘组织SGK1、TGF-β1阳性表达与胎盘组织胶原纤维沉积呈明显的正性相关(r=0.534,P<0.05,r=0.512,P<0.05)。结论子痫前期通过上调胎盘组织SGK1、TGF-β1阳性表达,从而加重胎盘组织胶原纤维沉积,导致疾病的发生与发展。
Objective To investigate the role of serum and glucocorticoid-induced protein kinase-1 (SGK1) and transforming growth factor β1 (TGF-β1) in the development and progression of preeclampsia in placenta. Methods From January 2014 to January 2015, 79 pregnant women with preeclampsia in our hospital were selected as the study group, including 39 cases of severe preeclampsia as group A and 40 cases of mild preeclampsia as group B, Forty normal pregnant women were used as the control group. The expression of SGK1 and TGF-β1 in the placenta was detected by immunohistochemical SP method. The placental collagen fibers were detected by Masson staining. The differences of SGK1 and TGF-β1 expression in placenta between groups were compared. Pearson correlation analysis was used to analyze the relationship between the expression of SGK1 and TGF-β1 in placenta and collagen deposition. Results The positive expression rate and positive expression of SGK1 and TGF-β1 in group A and group B were significantly higher than those in control group. The above indexes in group A and group SGK1 and TGF-β1 were significantly higher than those in group B (P <0.05). The deposition rate of collagen fibers in placenta of group A and group B was significantly higher than that of control group, and the above indexes in group A were significantly higher than those in group B, the difference was statistically significant (P <0.05). Pearson correlation analysis showed that positive expression of SGK1 and TGF-β1 in placenta was positively correlated with collagen deposition in placenta (r = 0.534, P <0.05, r = 0.512, P <0.05). Conclusion The positive expression of SGK1 and TGF-β1 in placental tissue of preeclampsia increased the deposition of collagen fibers in placenta, leading to the occurrence and development of the disease.
