目的 :研究维司力农结构改造物FPA对离体豚鼠心脏的正性肌力作用 ,探讨可能存在的抗心律失常作用及测定其急性毒性。方法 :豚鼠离体心脏实验采用Langendorff灌流法 ;急性毒性选用昆明种小白鼠进行实验 ,LD50 按Bliss法计算。结果 :在 0 5~ 2 0mM浓度范围内 ,FPA明显增加心脏收缩张力 ,1 0mM浓度时张力增幅达最大值。FPA各浓度组与阴性对照组 (0 4 %二甲基亚砜 )比较 ,心脏收缩张力增幅具有显著性差异 (P <0 0 1 ) ,而心率的负性变时效应无显著性差异 (P >0 0 5 )。 1 0mMFPA与阳性对照组 1 77× 1 0 -6M毒毛旋花甙K)比较 ,心脏收缩张力增幅不具有显著性差异 (P >0 0 5 ) ,而阳性对照组全部出现心律失常 ;FPA的LD50 为 2 5 1 0mg/Kg .结论 :FPA有较强的正性肌力作用 ,有一定的临床开发使用价值。
OBJECTIVE: To study the positive inotropic effect of FPA, a modified version of Verbil®, on the isolated guinea pig heart and to explore the possible anti-arrhythmic effects and to determine its acute toxicity. Methods: Langendorff perfusion method was used in guinea pig isolated heart experiment; Kunming mice were selected for acute toxicity test, LD50 was calculated by Bliss method. Results: In the concentration range of 0 5 ~ 20 mM, FPA significantly increased systolic tension and reached the maximum at 10 mM. Compared with the negative control group (0 4% dimethylsulfoxide), the FPA concentrations in each concentration group had significant differences in the increase of cardiac contractile tension (P <0.01), but no significant difference in the negative change of heart rate (P > 0 0 5). 1 0mMFPA and positive control group 1 77 × 1 0-6M tortoise velvet K) compared to the increase in cardiac contractility did not show significant difference (P gt; 0 05), while the positive control group all arrhythmias; FPA LD50 was 215 mg / kg.Conclusion: FPA has a strong positive inotropic effect, and has certain value in clinical development.
