目的探讨辐射对粘膜免疫的影响。方法小鼠全身照射,吸收剂量为3、8、12Gy,然后分离培养全小肠上皮内淋巴细胞(IEL),并对其数量、细胞增殖活力、细胞毒力、TNFα和TGFβ分泌水平进行检测。结果①全身照射后使IEL数量减少,3种剂量均呈现出伤后8小时下降,48~72小时达最低值,7天回升,至15天尚未恢复正常水平;各剂量组间差异无显著性,但8Gy组较3Gy组恢复慢,12Gy组未待回升动物已死亡;②全身照射后IEL功能受抑,表现为增殖活力和细胞毒力下降,其变化过程和数量减少相一致;③全身照射后IEL分泌TNFα和TGFβ增多,也呈现为8小时升高,48~72小时达高峰,15天仍未下降至正常值的过程。结论放射损伤导致IEL数量减少及功能降低,造成肠粘膜免疫功能的下降,从而共同导致肠道屏障功能的削弱
Objective To investigate the effects of radiation on mucosal immunity. Methods The whole body irradiation of mice was carried out, the absorbed dose was 3, 8 and 12 Gy, then the whole intestinal epithelial lymphocytes (IEL) were isolated and cultured, and their quantity, cell proliferation activity, cytotoxicity, TNFα and TGFβ secretion Test. Results ① The number of IEL decreased after the whole body irradiation. The three doses all showed a decrease of 8 hours after injury, reached the lowest value at 48-72 hours and rose at 7 days, and had not returned to normal level at 15 days. There was no significant difference among the three dosage groups , But the recovery of 8Gy group was slower than that of 3Gy group, and the animals of 12Gy group did not have to be resuscitated. (2) The IEL function was inhibited after systemic irradiation, which showed that the proliferation activity and cytotoxicity decreased, After IEL secretion of TNF α and TGF β increased, also showed an increase of 8 hours, 48 to 72 hours reached its peak, 15 days has not yet dropped to the normal process. Conclusions Radiation injury leads to the decrease of IEL quantity and function, resulting in the decrease of intestinal mucosal immune function, which leads to the impaired intestinal barrier function
