目的:探讨Castleman′s病患者外周血中CD4+CD25+调节性T细胞的变化及其与疾病的关系。方法:用流式细胞术对14例Castleman′s病患者及30例正常对照的外周血中CD4+CD25lowT细胞、CD4+CD25highT细胞、CD4+CD25highFOXP3+T细胞的比例进行检测。结果:局灶型Castleman′s病患者外周血中CD4+CD25lowT细胞、CD4+CD25highT细胞和CD4+CD25highFOXP3+T细胞比例在治疗前分别为(4.55±0.53)%、(0.73±0.09)%、(0.64±0.09)%;治疗后分别为(4.88±0.24)%、(1.0±0.10)%、(0.93±0.10)%,均低于正常对照组。治疗后这3种T细胞的比例较治疗前有显著升高(P<0.05)。多中心型患者外周血中这3种T细胞比例在治疗前分别为(9.43±0.81)%、(2.79±0.17)%、(2.46±0.09)%;治疗后(8.82±1.01)%、(2.71±0.10)%、(2.23±0.10)%,均高于正常对照组。结论:CD4+CD25+调节性T细胞在局灶型和多中心型Castleman′s病中的变化是不同的,这种不同的变化可能和这两型的临床表现和预后不同有关。
Objective: To investigate the changes of CD4 + CD25 + regulatory T cells in peripheral blood of patients with Castleman’s disease and their relationship with diseases. Methods: The proportion of CD4 + CD25lowT cells, CD4 + CD25highT cells and CD4 + CD25highFOXP3 + T cells in peripheral blood of 14 patients with Castleman’s disease and 30 normal controls were detected by flow cytometry. Results: The percentage of CD4 + CD25lowT, CD4 + CD25highT and CD4 + CD25high FOXP3 + T cells in peripheral blood of patients with Castleman’s disease were (4.55 ± 0.53)% and (0.73 ± 0.09)% (4.88 ± 0.24)%, (1.0 ± 0.10)% and (0.93 ± 0.10)%, respectively, which were lower than that of the normal control group. After treatment, the proportion of these three kinds of T cells was significantly higher than that before treatment (P <0.05). The proportions of these three kinds of T cells in the peripheral blood of patients with multi-center type were (9.43 ± 0.81)%, (2.79 ± 0.17)% and (2.46 ± 0.09)%, 8.82 ± 1.01% and 2.71 ± 0.10)%, (2.23 ± 0.10)%, respectively, which were higher than those in normal control group. CONCLUSION: The changes of CD4 + CD25 + regulatory T cells in focal and multicenter Castleman’s disease are different, and these different changes may be related to the different clinical manifestations and prognosis of these two types.
