目的:探讨顺铂抗人腺样囊性癌细胞(ACC-2)的作用机制。方法:采用MTT比色法研究顺铂对人腺样囊性癌(ACC-2)细胞和涎腺细胞(HSG)的相对选择性,应用原位杂交法检测人腺样囊性癌细胞端粒酶基因表达的变化。结果:顺铂对人腺样囊性癌细胞的抑制率高达98.4%,明显高于同期培养的涎腺细胞(HSG)(67.9%),具有显著性差异(P<0.01)。人腺样囊性癌细胞(ACC-2)的端粒酶阳性细胞为88.49%,顺铂组为25.62%,具有显著性差异(P<0.01)。结论:顺铂对人腺样囊性癌细胞(ACC-2)端粒酶基因的表达具有明显的抑制作用。
Objective: To investigate the mechanism of action of cisplatin on human adenoid cystic carcinoma cells (ACC-2). Methods: The relative selectivity of cisplatin on human adenoid cystic carcinoma (ACC-2) cells and salivary gland cells (HSG) was studied by MTT colorimetric assay. In situ hybridization was used to detect telomerase activity in human adenoid cystic carcinoma cells Changes in gene expression. Results: The inhibitory rate of cisplatin to human adenoid cystic carcinoma cells was as high as 98.4%, which was significantly higher than that of HSG cells (67.9%) in the same period (P <0.01). The telomerase positive cells in human adenoid cystic carcinoma cells (ACC-2) were 88.49% and 25.62% in cisplatin group (P <0.01). Conclusion: Cisplatin can significantly inhibit the telomerase gene expression in human adenoid cystic carcinoma cells (ACC-2).
