目的 培养狗肾细胞(MDCK)、人喉表皮癌细胞(HEP-2)、横纹肌肉瘤细胞(RD)混合细胞系(MHR),为检测呼吸道病毒和肠道病毒建立新方法.方法 将生长成片的MDCK、HEP-2、RD细胞按照1∶1∶1的比例混合,将甲型H1N1、H3N2、Victoria、Yamagata 4种亚型的流感病毒进行101~1010倍稀释,接种到MHR和MDCK中,在24 h、48 h、72 h、96 h、120h取培养物上清测血凝(HA)滴度;将肠道病毒CoxA16分别接种到MHR和RD、HEP-2细胞中.结果 甲型H1N1、H3N2、Victoria、Yamagata 4种亚型流感病毒在MDCK细胞的血凝滴度高于或等于混合细胞,能检测到的最低浓度高于混合细胞,CoxA16病毒在3种细胞出现CPE的时间长短为RD< MHR< HEP-2.结论 MHR对流感病毒和肠道CoxA16病毒均较敏感,可用于流感病毒和肠道病毒的分离培养.“,”Objective To establish a new method for detecting respiratory viruses and enteroviruses by using mixed cell(MHR) with canine kidney cell (MDCK),human epidermoid cancer cells(HEP-2) and rhabdomyosarcoma cell (RD).Methods MDCK,HEP-2,RD cells were mixed according to the ratio of 1 ∶ 1 ∶ 1,four subtypes of H1N1,H3N2,Victoria and Yamagata,influenza virus were 101-1010 times diluted and culture in MHR mixed cell and MDCK cell.We measured hemagglutination titer of cultured supernatant measured in 24 h,48 h,72 h,96 h,120 h.Enteroviruses virus CoxA16 was planted with MHR,HEP-2 and RD cell.Results The HA of four kinds of subtypes influenza virus H1 N1,H3N2,Victoria,Yamagata was higher or equal in MDCK cell than in the MHR mixed cell,and the lowest detected concentration was higher than that of MDH mixed cells,time of the CPE CoxA16 virus in cells followed the order of RD < MHR < HEP-2.Conclusion MHR mixed cells is sensitive to influenza virus and enteroviruse CoxA16 virus and can be used for the isolation of influenza virus and enterovirus.
