目的本研究在中国浙东地区汉族人群中,探讨氧化低密度脂蛋白受体基因单核苷酸多态性与冠心病患病风险的关联性。方法对159例冠心病患者和106例非冠心病者样本采用4管SYBR-PCR结合融解曲线方法进行G501C等位基因型的检测,进而作出基因诊断。随机选取50份经荧光定量PCR分型的标本进行DNA测序鉴定。结果 4号外显子上的G501C基因多态性在冠心病患者组和对照组中的基因型分布和等位基因频率差异无统计学意义(P>0.05)。经过荟萃分析,也没有发现G501C与CAD的关联性,结论 G501C基因位点多态性可能与浙东地区汉族人群CAD的发病无相关性,不能作为冠心病的遗传易感标志。
Objective To investigate the association between single nucleotide polymorphism of oxidized low density lipoprotein receptor gene and the risk of coronary heart disease in Han population of eastern Zhejiang, China. Methods A total of 159 coronary heart disease patients and 106 non-coronary heart disease patients were tested for G501C allele by 4-SYBR-PCR combined with thawing method. 50 randomly selected fluorescence quantitative PCR typing of specimens for DNA sequencing. Results There was no significant difference in the distribution of genotypes and allele frequencies of G501C polymorphism in Exon 4 between CHD patients and controls (P> 0.05). After meta-analysis, there was no correlation between G501C and CAD. Conclusion The polymorphism of G501C locus may not be associated with CAD in Han population of Eastern Zhejiang Province, and can not be used as a genetic susceptible marker of coronary heart disease.
