目的 研究氧化苦参碱与国产α 1b -和α 2b -干扰素对大鼠肝星状细胞增殖和胶原合成的影响 ,为应用氧化苦参碱防治肝纤维化提供理论依据。方法 分离、培养大鼠肝星状细胞 ,采用MTT法和3 H -脯氨酸掺入法分别测定细胞增殖和胶原合成。结果 氧化苦参碱在 5 μg~ 5 0 0 μg/mL ,两种α -干扰素在 5 0 0U -10 0 0 0U/mL浓度范围内能呈剂量依赖性抑制星状细胞的增殖和胶原合成 ,提高药物浓度则形成作用平台。结论 氧化苦参碱与α -干扰素在体外均具有抑制大鼠肝星状细胞增殖和胶原合成的作用 ,这对防治肝纤维化可能有一定的临床应用价值
Objective To investigate the effects of oxymatrine and domestic α 1b - and α 2b - interferon on the proliferation and collagen synthesis of rat hepatic stellate cells and provide a theoretical basis for the prevention and treatment of hepatic fibrosis with oxymatrine. Methods Rat hepatic stellate cells were isolated and cultured. Cell proliferation and collagen synthesis were determined by MTT assay and 3 H - proline incorporation assay. Results Oxymatrine inhibited the proliferation and collagen synthesis of stellate cells in a dose - dependent manner at a concentration of 5 000 ~ 100 000 μg / mL. , Increase the concentration of drugs to form a role platform. Conclusion Both oxymatrine and α - interferon can inhibit rat hepatic stellate cell proliferation and collagen synthesis in vitro, which may have some clinical value in prevention and treatment of liver fibrosis
