人骨髓来源的间充质干细胞(BMSCs)具有很重要的临床治疗价值,小鼠BMSCs为研究观察这些前体细胞群的生物学性能提供了重要的基础模型。本研究从转基因小鼠骨髓中早期分离培养BMSCs,并进行鉴定。把Twist2-cre小鼠与含有Cre报告子的小鼠(如Z/EG和Ai9)杂交,可分别表达EGFP和番茄红荧光蛋白,其中的Cre可切除终止子序列。运用该方法在细胞克隆形成最早期,筛选出一种能表达荧光蛋白的T2C+细胞群,培养到第5天时该群细胞约占总细胞群的20%。
Human bone marrow-derived mesenchymal stem cells (BMSCs) have important clinical value. The mouse BMSCs provide an important basic model for studying the biological properties of these precursor cell populations. In this study, BMSCs were isolated from bone marrow of transgenic mice and identified. Twist2-cre mice were crossed with mice containing the Cre reporter, such as Z / EG and Ai9, to express EGFP and tomato red fluorescent protein, respectively, where Cre excises the terminator sequence. Using this method, at the earliest stage of cell cloning, a T2C + cell population capable of expressing the fluorescent protein was screened, and on day 5, the group of cells accounted for about 20% of the total cell population.
