首先将刀豆蛋白A(ConA)交联于溴化氢活化的Sepharose 4B上,与胃癌物组织可溶性提取液充分反应后,用亲和层析的方法纯化出ConA结合物,经甲醇氯仿抽提其糖脂成分行高效薄层色谱分析,证实胃癌组织ConA结合的糖脂主要是一种糖链较长的中性糖脂。这对于胃癌组织中ConA受体表达机理的探讨及其在诊断方面的应用都具有重要意义,同时也建立了一种利用ConA亲和层析分离提取胃癌中性糖脂的方法。
Firstly, ConA was cross-linked to hydrogen bromide-activated Sepharose 4B and fully reacted with the soluble extract of gastric cancer tissue. ConA conjugate was purified by affinity chromatography and extracted with methanol-chloroform Its glycolipid components were analyzed by high performance thin layer chromatography, confirming that ConA-bound glycolipid in gastric cancer tissue is mainly a kind of neutral glycolipid with longer sugar chain. This is of great significance for the study of the mechanism of ConA receptor expression in gastric cancer and its application in diagnosis. Meanwhile, a method for the separation and extraction of gastric cancer glycolipid by ConA affinity chromatography has also been established.
