目的探讨逍遥散对慢性应激模型大鼠血清肿瘤标志物的影响。方法 Wistar大鼠30只,随机分作模型组、空白组和治疗组,常规饲养,模型组在常规饲养基础上另外给予对应的慢性应激刺激,治疗组在模型组处理基础上给予中药逍遥散煎剂灌胃,21天后采血清,Elisa法测定血清CEA、CA19-9、CA724、AFP。统计学处理,用线性回归分析肿瘤标志物与慢性应激的相关性,方差分析比较三组血清肿瘤标志物差异。结果肿瘤标志物测定值如下,空白组:CEA:9.07±1.43,CA19-9:321.01±35.95,CA724:8.43±1.02,AFP:7.20±0.80;模型组:CEA:22.73±1.6,CA19-9:729.11±57.68,CA724:18.66±1.11,AFP:15.83±0.89;治疗组:CA19-9:521.16±34.73,CA724:12.91±1.18,AFP:7.63±1.20。回归分析提示CA19-9、CA724、AFP与慢性应激有相关性,且以AFP相关性最高。方差分析三组间比较,CA19-9、CA724、AFP三组间差异均有统计学意义(P<0.01);LCE-T检验两两组间比较,CA19-9、CA724治疗组与空白组,治疗组与模型组,模型组与空白组之间差异均有统计学意义(P<0.01),AFP治疗组与模型组,模型组与空白组之间差异均有统计学意义(P<0.01),治疗组与空白组之间比较差异无统计学意义(P>0.05)。结论慢性应激损伤能刺激大鼠血清肿瘤标志物升高,有直接诱发肿瘤发生的可能性。慢性应激可能与消化系统肿瘤(特别是肝癌)的发生,关系较为密切。中药方剂逍遥散能够有效降低慢性应激诱发的肿瘤标志物升高,有望预防慢性应激诱发的癌变。
Objective To investigate the effect of Xiaoyaosan on serum tumor markers in chronic stress rats. Methods Thirty Wistar rats were randomly divided into three groups: model group, blank group and treatment group. The rats in the model group were given conventional chow diet. Chronic stress stimulation was given to the model group on the basis of routine feeding. The treatment group was given Xiaoyao San Decoction gavage, 21 days after the sera, Elisa method for the determination of serum CEA, CA19-9, CA724, AFP. Statistical analysis, using linear regression analysis of tumor markers and chronic stress correlation analysis of variance three groups of serum tumor markers. Results The tumor markers were as follows: blank group: CEA: 9.07 ± 1.43, CA19-9: 321.01 ± 35.95, CA724: 8.43 ± 1.02, AFP: 7.20 ± 0.80; model group: CEA: 22.73 ± 1.6, CA19-9: 729.11 ± 57.68, CA724: 18.66 ± 1.11, AFP: 15.83 ± 0.89; treatment group: CA19-9: 521.16 ± 34.73, CA724: 12.91 ± 1.18, AFP: 7.63 ± 1.20. Regression analysis showed that CA19-9, CA724, AFP correlated with chronic stress, and had the highest correlation with AFP. There were significant differences among the three groups in CA19-9, CA724 and AFP among the three groups (P <0.01). There was significant difference between the two groups in CA19-9, CA724 treatment group and LCE-T test The differences between the AFP treatment group and the model group, the model group and the blank group were statistically significant (P <0.01). There were significant differences between the AFP treatment group and the model group, the model group and the blank group (P <0.01) There was no significant difference between the treatment group and the blank group (P> 0.05). Conclusion Chronic stress can stimulate the increase of serum tumor markers in rats, which may directly induce the tumorigenesis. Chronic stress may be related to the occurrence of digestive system tumors (especially liver cancer), which are closely related. Chinese medicine Xiaoyaosan can effectively reduce the chronic stress-induced tumor markers increased, is expected to prevent chronic stress-induced carcinogenesis.
