化性是家蚕的重要生理特性,受第6染色体上的化性主基因V及Z染色体上的伴性成熟基因Lm的影响,伴性早熟基因Lme促进家蚕化性趋向多化性.采用多化性家蚕品种nistari和二化性家蚕品种p50配制BC1M群体[p50×(nistari×p50)],选择其中734头产非滞育卵的雌蛾个体作为定位群体,以其基因组DNA为模板进行SSR标记的PCR扩增,应用高分辨率熔解曲线分析(HRM)技术进行各个标记位点的基因型分析,绘制出遗传总距离为12.91 cM的分子遗传连锁图,Lme基因位于标记s2734-299和s2734-525之间,与2个标记的遗传距离均为0.41 cM.s2734-299和s2734-525间的物理距离为226 kb,其间有18个预测基因,有2个属于DNA序列特异性结合转录因子基因,但尚不能判定这2个基因是否与Lme有明确的关联.“,”Voltinism is an important physiological characteristic of silkworm (Bombyx mori),which is affected by the dominant V gene located on chromosome 6 and the sex-linked maturation gene (Lm) located on sex chromosome Z.The sexlinked early maturation gene (Lme) could promote change of silkworm voltinism towards multivoltinism.Here,silkworm multivoltine variety nistari and bivoltine variety p50 were used as parental strains to construct BC1M population [p50×(nistari× p50)],among which 734 female moths laying non-diapause eggs were selected as mapping group for PCR amplification of their genomic DNAs.SSR molecular markers were genotyped by high-resolution melting (HRM) analysis technique.A map covered a total genetic distance of 12.91 cM was obtained.Lme was located between markers S2734-299 and S734-525,and the genetic distances between Lme and these two markers was both O.41 cM.Physical distance between the two markers was 226 kb.There were altogether 18 predicted genes in this region.However,two genes coding for DNA-binding specific transcription factors cannot be determined as associated genes to Lme.
