目的探讨钙磷涂层对兔下颌骨镁合金植入体周围软组织中Ⅰ型胶原mRNA表达的影响。方法本研究于2008年12月至2009年8月在中国医科大学实验动物中心和中心实验室进行。选择雄性成年日本大耳白兔27只,随机分为T组、A组和B组,每组9只。将钙磷涂层镁合金螺钉(T组)和无处理镁合金螺钉(A组)植入兔的下颌骨内,以钛合金(B组)作为对照组,每组观察时间分别为1、2、3个月。采用反转录聚合酶链反应(RT-PCR)方法检测植入体周围软组织中Ⅰ型胶原的mRNA表达情况。结果 T组Ⅰ型胶原的mRNA表达水平高于A组,随观察时间的增加,两组Ⅰ型胶原的mRNA表达量均逐渐增高。观察2和3个月时的组间差异有统计学意义(P<0.05),观察1个月时的组间差异无统计学意义(P>0.05);T组观察2和3个月的Ⅰ型胶原mRNA表达水平与1个月相比,均有统计学意义(P<0.05)。结论钙磷涂层能够提高镁合金植入体周围软组织Ⅰ型胶原的mRNA表达水平,促进早期的成骨过程,显示出良好的生物活性。
Objective To investigate the effect of calcium phosphate coating on the expression of type Ⅰ collagen mRNA in soft tissues around the mandibular magnesium alloy implant. Methods The study was performed at Experimental Animal Center and Central Laboratory, China Medical University from December 2008 to August 2009. Twenty-seven male Japanese white rabbits were randomly divided into T group, A group and B group, with 9 in each group. Titanium alloy (group B) was used as the control group, and the observation time of each group was 1, 2 ,3 months. Reverse transcription polymerase chain reaction (RT-PCR) was used to detect mRNA expression of type I collagen in peri-implant soft tissues. Results The mRNA expression of type Ⅰ collagen in group T was higher than that in group A, and the mRNA expression of type Ⅰ collagen in both groups gradually increased with the increase of observation time. The difference between the two groups was statistically significant at 2 and 3 months (P <0.05). There was no significant difference between the two groups at 1 month (P> 0.05) Collagen mRNA expression level compared with 1 month, were statistically significant (P <0.05). Conclusion Calcium-phosphorus coating can improve the mRNA expression of type Ⅰ collagen in soft tissue around the magnesium alloy implant, promote early osteogenesis and show good biological activity.
