用粘附式细胞仪(ACAS570)检测了培养细胞缝隙连接通讯功能。预先用荧光探针6-羟基荧光素乙酰乙酸盐进行细胞染色,然后光漂白细胞中的荧光分子。由于邻近细胞中未漂白的荧光分子通过缝隙连接扩散到漂白的细胞内,故可监测漂白细胞荧光强度的恢复。根据荧光恢复的比率,了解缝隙连接通讯功能的状态。
Cell-gap junctional communication was examined using an adherent cytometer (ACAS570). Cells were stained with the fluorescent probe 6-hydroxyfluorescein acetoacetate in advance and then photobleached the fluorescent molecules in the cells. Since the unbleached fluorescent molecules in adjacent cells diffuse into the bleached cells through gap junctions, the recovery of the fluorescent intensity of the bleached cells can be monitored. According to the ratio of fluorescence recovery, understand the state of the gap connection communication function.
