目的:研究辛润苦泄法对胰岛素抵抗大鼠肝脏和脂肪组织中肿瘤坏死因子-α(TNF-α)和过氧化物酶体增殖物激活受体-γ(PPAR-γ)表达的影响。方法:采用高糖高脂饲料喂养制作胰岛素抵抗大鼠模型,将成功造模的30只大鼠随机分为模型组、唐旨宁组和文迪雅组,每组10只。药物干预6周后,用ELISA方法定量测定血清中TNF-α水平;免疫组织化学方法检测肝脏和脂肪组织中PPAR-γ蛋白表达;RT-PCR法检测肝脏和脂肪组织中TNF-αmRNA及PPAR-γmRNA的表达。结果:高糖高脂饲料喂养6周后,模型组大鼠空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)和血清胰岛素(FIN)s均明显高于对照组(P<0.01),胰岛素敏感性指数(ISI)明显低于对照组(P<0.01);药物干预6周后,与模型组比较,唐旨宁组和文迪雅组大鼠血清TNF-α显著降低(P<0.01),肝脏和脂肪组织中PPAR-γ蛋白的表达均显著增高(P<0.01,P<0.05),TNF-αmRNA表达明显减少(P<0.05,P<0.01),PPAR-γmRNA表达显著增加(P<0.01)。结论:唐旨宁能明显改善胰岛素抵抗大鼠的糖脂毒性,其作用机制可能是通过下调TNF-α、上调PPAR-γ的表达而实现的。
Objective: To study the effect of Xinrun Kuxie Method on the expression of tumor necrosis factor-α (TNF-α) and peroxisome proliferator-activated receptor-γ (PPAR-γ) in the liver and adipose tissue of insulin resistance rats. Methods: A rat model of insulin resistance was established by feeding with high-sugar and high-fat diet. Thirty rats were randomly divided into model group, Tang Zuening group and Avandia group, with 10 rats in each group. Serum levels of TNF-α were measured by enzyme-linked immunosorbent assay (ELISA). PPAR-γ protein expression in liver and adipose tissue was detected by immunohistochemical method. TNF-α mRNA and PPAR- γ mRNA expression. Results: Fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG) and serum insulin (FIN) s in the model group were significantly higher than those in the control group after 6 weeks of high glucose and high fat diet (P < 0.01). The ISI was significantly lower than that of the control group (P <0.01). Compared with the model group, the levels of serum TNF-α in the Tangzhining and Avandia groups were significantly decreased (P <0.01, P <0.05). The expression of PPAR-γmRNA in liver and adipose tissue were significantly increased (P <0.01, P <0.05) (P <0.01). CONCLUSION: Tang Xuning can significantly improve the glycolipid toxicity of insulin resistance rats, and its mechanism may be through down-regulating TNF-α and up-regulating the expression of PPAR-γ.
