目的:研究桂枝茯苓丸(GFW)对S180荷瘤小鼠移植瘤生长和转移的抑制作用,探讨其作用机制。方法:复制S180荷瘤小鼠模型,计算抑瘤率。采用SABC免疫组化法检测肿瘤细胞中细胞周期调控基因p53及细胞黏附分子CD44表达。结果:桂枝茯苓丸有抑瘤作用,抑瘤率为33.48%。GFW与环磷酰胺(CY)合用抑瘤作用显著提高,抑瘤率为57.17%。桂枝茯苓丸可下调肿瘤细胞突变型p53蛋白的表达,与模型组比较,差异有统计学意义(P<0.01)。且可抑制细胞黏附分子CD44的表达,与模型组比较,差异有统计学意义(P<0.05)。结论:桂枝茯苓丸具有明显抑瘤作用,且对化疗药物环磷酰胺抗S180肉瘤具有增效作用。其抑瘤机制可能是GFW影响肿瘤细胞细胞周期调控基因p53和细胞黏附分子CD44的表达,即通过影响肿瘤细胞增殖周期,降低肿瘤细胞与基质之间的黏附,从而达到抑制肿瘤细胞生长和转移的作用。
Objective: To study the inhibitory effect of Guizhi Fuling Capsule (GFW) on the growth and metastasis of S180 tumor-bearing mice xenografts and to explore its mechanism. Methods: S180 tumor-bearing mouse model was duplicated and the tumor inhibition rate was calculated. SABC immunohistochemistry was used to detect the expression of cell cycle regulatory gene p53 and cell adhesion molecule CD44 in tumor cells. Results: Guizhi Fuling pill has anti-tumor effect, inhibition rate was 33.48%. The combination of GFW and cyclophosphamide (CY) significantly increased the antitumor activity, inhibition rate was 57.17%. Guizhi Fuling pill can down-regulate the expression of mutant p53 protein in tumor cells, which has statistical significance compared with model group (P <0.01). And inhibited the expression of CD44. Compared with the model group, the difference was statistically significant (P <0.05). Conclusion: Guizhi Fuling pill has obviously anti-tumor effect, and has synergistic effect on chemotherapy drug cyclophosphamide anti-S180 sarcoma. The anti-tumor mechanism may be GFW affect the tumor cell cycle control gene p53 and cell adhesion molecule CD44 expression, that is, by affecting the tumor cell proliferation cycle, reducing the adhesion between tumor cells and matrix to achieve inhibition of tumor cell growth and metastasis effect.
