目的:观察中药复方消斑通脉合剂药物血清对体外培养的VSMCs增殖的作用。方法:体外培养VSMCs,取对数生长期细胞用作实验,MTT法检测不同浓度药物血清对体外培养的VSMCs增殖活力,免疫组织化学方法检测各组药物血清对VSMCs中P21表达的影响。结果:MTT法显示药物血清呈剂量依赖性抑制VSMCs增值,高剂量与低剂量比较有统计学差异(P<0.05),免疫组化显示药物血清可明显促进VSMCs中P21表达,高剂量与低剂量比较有统计学差异(P<0.05)。结论:中药复方消斑通脉合剂药物血清通过促进VSMCs中P21表达抑制VSMCs增殖。
Objective: To observe the effect of traditional Chinese medicine Xiaobantongmai mixture on the proliferation of cultured VSMCs in vitro. Methods: VSMCs were cultured in vitro. The cells in logarithmic growth phase were used as the experiment. The proliferative activity of VSMCs cultured in vitro was assayed by MTT assay. The effect of serum of each group on the expression of P21 in VSMCs was detected by immunohistochemistry. Results: MTT assay showed that serum of the drug increased the proliferation of VSMCs in a dose-dependent manner. There was a significant difference between the high dose and the low dose (P <0.05). Immunohistochemistry showed that serum of the drug significantly promoted P21 expression in VSMCs. There was a statistically significant difference (P <0.05). Conclusion: The Chinese medicine compound Xiaobantongmai Mixture can inhibit the proliferation of VSMCs by promoting the expression of P21 in VSMCs.
