目的:制备平阳霉素壳聚糖温度敏感型原位凝胶,并进行流变学性质研究及体外评价。方法:采用流变仪考察平阳霉素原位凝胶体系的流变学性质。通过体外释放及体外降解研究,对制剂进行体外评价。结果:流变学研究表明,随着温度升高到37℃左右时,原位凝胶溶液发生了从溶液向凝胶的相转变过程。体外释药的研究表明,药物能持续释放10 d以上,累积释放量达到85%以上。结论:以上结果表明平阳霉素原位凝胶符合介入栓塞要求,体外具有明显缓释效果。
OBJECTIVE: To prepare temperature-sensitive in situ gels of pingyangmycin chitosan and study its rheological properties and evaluation in vitro. Methods: The rheological properties of pingyangmycin in situ gel system were investigated by rheometer. The formulations were evaluated in vitro by in vitro release and in vitro degradation studies. Results: Rheological studies showed that phase transition from solution to gel occurred in situ gel solution as the temperature increased to about 37 ° C. In vitro drug release studies show that the drug can be sustained release of more than 10 d, the cumulative release of more than 85%. Conclusion: The above results show that bleomycin in situ gel in line with the requirements of intervention embolism, in vitro has a significant sustained-release effect.
