物种的DNA C-值具有重要的生态意义,同时也具有种的特征,目前DNA C-值越来越广泛地应用于物种倍性和杂种鉴定、植物生态适应能力分析等。本文应用流式细胞术,以油菜(Brassica campes-tris)为材料,从标样、细胞核悬液、样品生长时期等方面研究了影响DNA C-值检测的实验因素,发现标样种类和样品的生长时期对实验结果没有明显影响;与Galbraith缓冲液和Otto缓冲液相比,应用Tris-MgCl2细胞核悬液能够得到稳定的实验结果。
DNA C-value of species has important ecological significance, but also has the characteristics of species, the current DNA C-value is more widely used in species ploidy and hybrid identification, plant ecological adaptability analysis. In this paper, we used flow cytometry and Brassica campestris as the material to study the experimental factors that influence the DNA C-value detection from the standard sample, the nucleus suspension, the sample growth period and so on, and found that the sample type and the sample The growth period had no significant effect on the experimental results; stable experimental results were obtained using Tris-MgCl2 nuclear suspensions compared to Galbraith’s buffer and Otto’s buffer.
