目的研究高良姜素促进人乳腺癌细胞株MCF-7的凋亡作用。方法采用CCK-8法检测高良姜素对MCF-7细胞的增殖抑制作用;以流式细胞术检测细胞凋亡;Western blot法检测Bax、Bcl-2、cleavedcaspase-3和cleaved-PARP等相关蛋白的表达水平。结果高良姜素对MCF-7细胞增殖有明显的抑制作用,24 h和48 h的IC50分别为43.71μmol·L~(-1)和20.68μmol·L~(-1);高良姜素能够诱导MCF-7细胞凋亡,呈浓度依赖关系;高良姜素能上调Bax、cleaved-caspase-3和cleaved-PARP蛋白的表达,下调Bcl-2蛋白的表达。结论高良姜素对MCF-7细胞增殖有明显的抑制作用,通过线粒体途径调节凋亡相关蛋白的表达水平诱导MCF-7细胞凋亡。
Objective To investigate the effects of galangin on the apoptosis of human breast cancer cell line MCF-7. Methods The inhibitory effect of galangin on the proliferation of MCF-7 cells was detected by CCK-8 assay. The apoptosis of MCF-7 cells was detected by flow cytometry. The expressions of Bax, Bcl-2, cleaved caspase-3 and cleaved-PARP were detected by Western blot The level of expression. Results Galangin could significantly inhibit the proliferation of MCF-7 cells, and the IC50 values were 43.71μmol·L -1 and 20.68μmol·L -1 at 24 and 48 h, respectively. Galangin was able to induce The apoptosis of MCF-7 cells in a concentration-dependent manner; galangin can up-regulate Bax, cleaved-caspase-3 and cleaved-PARP protein expression, down-regulate the expression of Bcl-2 protein. CONCLUSION Galangin can significantly inhibit the proliferation of MCF-7 cells and induce the apoptosis of MCF-7 cells by regulating the expression of apoptosis-related proteins through mitochondrial pathway.
