大白鼠体内碳氧血红蛋白生物半衰期初探

一氧化碳(CO)是目前影响人类健康的主要空气污染物和空气污染监测指标之一.CO进入人体后的主要存在形式碳氧血红蛋白(HbCO)的生物半衰期在128~409min之间,但动物体内HbCO的消长规律报道很少.为便于用动物研究CO的毒性,我们对大白鼠体内HbCO的生物半衰期进行了实验研究,现将结果报告如下.1 材料与方法1.1 CO来源 以普通蜂窝煤炉作为CO发生源.1.2 动物选择 取健康大白鼠雌雄各4只.体重范围雌性在 240.0~278 og之间,雄性在 310.0~457.5g之间.1.3 实验方法 将大白鼠置室外暴露2h之后,室外CO浓度为3.71mg/m~3,采尾血20μl和40μl分别测定Hb和HbCO含量.然后将其移入容积为38.73m~3的染毒室内染毒30min,染毒室内平均CO浓度为349.25mg/m~3.在停止接触后的0、30、60、90、120min分别采尾血40μl,用CO-I型一氧化碳测定仪测定血样与浓硫酸作用释出的CO量,进而计算CO含量.
