目的:探讨白细胞在早期糖尿病大鼠视网膜微血管中粘附、堆积,以致引起微循环障碍的作用。方法:健康成年雄性Wistar大鼠16只,随机分成正常对照组和链尿佐菌素(Streptozotocin,STZ)糖尿病3个月组各8只;对其视网膜消化铺片进行形态学观察及白细胞共有抗原(CD45)单克隆抗体免疫组织化学研究。结果:发现3个月病程的STZ诱导的糖尿病大鼠视网膜毛细血管已出现毛细血管管径不规则,呈节段性膨大或囊样扩张等形态学改变;视网膜毛细血管中CD45的表达明显增加,白细胞呈串珠样粘附和堆积。结论:3个月病程的STZ糖尿病大鼠可出现视网膜微血管改变;白细胞在糖尿病视网膜病变的发生发展中起着重要作用。
Objective: To investigate the leukocyte adhesion and accumulation in retinal microvessels of early diabetic rats, which may cause microcirculation disturbance. Methods: Sixteen healthy adult male Wistar rats were randomly divided into normal control group and streptozotocin (STZ) diabetic group for 8 months for 8 months. Morphological observation of the retina digesting and leukocyte common antigen (CD45) monoclonal antibody immunohistochemistry. Results: The retinal capillaries of STZ-induced diabetic rats of 3 months duration were found to have irregular capillary vessels with morphological changes such as segmental enlargement or cystic dilatation. The expression of CD45 in retinal capillaries was significantly increased, Leukocytes are beaded like a sticky and stacked. CONCLUSION: Retinal microvascular changes may occur in STZ diabetic rats of 3 months duration. Leukocytes play an important role in the development of diabetic retinopathy.
