目的观察金雀异黄素在低雌激素水平情况下对雌激素依赖阳性人乳腺癌细胞MCF-7的生长影响。方法采用四甲基偶氮噻唑蓝(MTT)法检测细胞增殖,流式细胞术分析细胞周期分布情况,磷脂结合蛋白V/碘化丙啶(AnnexinV/PI)联合双染法检测细胞凋亡,间接免疫荧光法测定雌激素调节蛋白(presenilin2,PS2)的表达。结果当MCF-7细胞经过去雌激素处理以后,与对照组相比,金雀异黄素在5×10-7~10-5mol/L时可以促进细胞增殖,G0/G1期细胞向S期推进,PS2蛋白表达增加,但是抑制细胞凋亡作用并不明显。结论金雀异黄素在低雌激素水平情况下可以促进人乳腺癌细胞MCF-7的细胞增殖、DNA合成以及PS2蛋白的表达,具有一定的雌激素样作用。
Objective To observe the effect of genistein on the growth of estrogen-dependent human breast cancer cell line MCF-7 under low estrogen levels. Methods Cell proliferation was detected by MTT assay. Cell cycle distribution was analyzed by flow cytometry. Apoptosis was detected by Annexin V / PI staining and double staining. Indirect immunofluorescence assay was used to determine the expression of presenilin2 (PS2). Results When MCF-7 cells were treated with de-estrogen, genistein could promote cell proliferation when treated with 5 × 10-7 ~ 10-5mol / L genistein, and cells in G0 / G1 phase increased to S phase Promote, PS2 protein expression increased, but the inhibition of apoptosis is not obvious. Conclusions Genistein can promote cell proliferation, DNA synthesis and expression of PS2 protein in human breast cancer cell line MCF-7 under low estrogen level, and has certain estrogen-like effects.
